746 Transcriptomic profiling of frontal and occipital dermal papilla reveals potential role of TRPS1 in androgenic alopecia

In androgenic alopecia (AGA), hair follicles in the balding scalp miniaturize. This is accompanied by shortened anagen, premature catagen and longer telogen. There are no changes to bulge cells (BC) AGA but there a decrease progenitor cells. We hypothesize loss of progenitors due aberrant signaling from dermal papilla (DP) scalp, which important for BC activation anagen induction. could increase telogen phase may contribute miniaturization. To determine if differences initiation ability exists between (frontal, FDP) nonbalding (occipital, ODP) DP, we developed an vitro double spheres assay that combined FDP or ODP with BC. tested induction marker keratin 75 (K75) found induce 1.23X reduction (p<0.01) K75 expression compared ODP, suggesting impaired differentiation FDP. undertook transcriptomic analysis intact find this upregulation TRPS1 1.7X cultured spheres, difference remained significant reduced 1.2X.
